KST 513: AGRICULTURAL BIOTECHNOLOGY
DATE: THURSDAY, 3RD MAY 2012 TIME: 2.00 P.M. – 5.00 P.M.
INSTRUCTIONS: Answer any four questions.
1. Write short notes on: (5 marks each)
i) Applications of embryo culture
ii) Mutation induction
iii) Purines and pyrimidines
iv) Processing of mRNA
v) Dominant and codominant DNA markers
2. Murashige and Skoog (MS) stock solutions have been prepared as indicated below:
(i) Macronutrients (x 10)
(ii) Micronutrients (x100)
(iii) Amino acids (x 1000)
(iv) Vitamins (x 1000)
(a) Indicate how much solution (mls) should be drawn from the stock solution to
make five litres of MS media. (8 marks)
(b) Two percent sucrose and 0.8% agar is added to the media above.
Calculate the weights of sucrose and agar added in five-litre media. (10 marks)
(c) Six (6) parts per million BA is added to the media above. Calculate weight of BA added to the media in grams. (7 marks)
Suppose stock solutions have been prepared as follows (25 marks):
(a) BA is 25 mg per 100ml
(b) NAA is 20 mg per 200ml
Calculate volume of stock solutions to make five litres medium with 4 mg per litre BA and 0.2 mg per litre NAA.
4. Discuss application of ten biotechnology tools in crop improvement. (25 marks)
5. A number of issues on GMOs have been resolved. Discuss at least five. (25 marks)
